Structural genomic variation in yeast diploids: Mechanisms and phenotypic consequences

The JL Argueso research program focuses on the structural plasticity of chromosomes. Specifically, we are interested in the endogenous and environmental factors that disrupt chromosomal integrity, and the cellular mechanisms that counteract these forces by repairing broken DNA. To address a variety of questions in structural genomic variation, my research takes advantage of the unmatched genomic investigation tools available in the Saccharomyces cerevisiae model. Our group has developed powerful cell-based assay systems to detect, quantify and characterize chromosomal rearrangements of both mitotic and meiotic origins leading to copy number variation (CNV), as well as mitotic recombination leading to Loss-of-Heterozygosis (LOH). In addition, we have recently expanded the scope of our structural variation studies to analyze whole chromosome gain and loss events (aneuploidy). We are using these innovative assays, in combination with whole genome analysis approaches, to dissect the molecular mechanisms of structural variation, and to identify environmental exposures that can elevate the basal rate of these chromosome-scale mutations.
