Deciphering the regulatory language of RNA localization
Thousands of RNAs have been observed to be asymmetrically localized within the cells of various cell types, tissues, and species. However, the molecular mechanisms that govern their intracellular transport are known for only a small handful. Given that defects in RNA transport are associated with a range of human neurological diseases, this knowledge gap limits our ability to explain observed phenotypes and design effective therapies. To confront this, our laboratory uses genome engineering, subcellular transcriptome sequencing, and single molecule imaging to uncover RNA cis-elements and their cognate RNA binding-proteins that dictate where RNAs end up within cells.
