Gsk-3-Dependent Signaling Regulates m6A mRNA Methylation and Promotes Pluripotency in Mouse Embryonic Stem Cells. 

Despite the many uses and recognized importance of pluripotent embryonic stem cells (ESCs), the biochemical pathways and genetic stimuli that influence the maintenance of ESC pluripotency are poorly understood. The insulin/PI3K pathway and reduced m6A RNA methylation are well known regulators of ESC pluripotency and a common link between these regulators is glycogen synthase kinase-3 (Gsk-3). The role of Gsk-3 in the insulin/PI3K pathway is well-established, while the influence of Gsk-3 in m6A methylation was recently discovered by our lab. A knowledge gap remains, however, with regards to ESC pluripotency regulation and whether Gsk-3-dependent signal transduction regulates RNA methylation in maintaining ESC pluripotency.
